Basic Research for the Future of BNCT

Basic research forms the foundation for the future success of Boron Neutron Capture Therapy (BNCT). No matter how advanced the treatment system may be, BNCT cannot achieve its full therapeutic potential unless boron-containing drugs accumulate selectively in tumor cells. Conversely, excessive uptake of boron by normal tissues limits the safety of the treatment.

At the Fujita Health University BNCT Research Center, we investigate the fundamental scientific questions that underpin successful BNCT. Our research includes the development of boron compounds with improved tumor selectivity, evaluation of their biodistribution, visualization using positron emission tomography (PET), optimization of neutron irradiation protocols, establishment of dose constraints for normal organs, and strategies to deliver neutrons more uniformly to deep-seated tumors.

One of the greatest challenges in BNCT is the treatment of deep-seated cancers. To achieve successful therapy, sufficient neutrons must reach tumors located deep within the body while minimizing radiation exposure to surrounding healthy organs, such as the gastrointestinal tract. Overcoming this challenge is essential for expanding the clinical application of BNCT.

In addition, our research focuses on cancer-specific molecular targets such as the L-type amino acid transporter 1 (LAT1). Understanding the unique biological characteristics of cancer cells may lead to more selective boron delivery systems, the development of novel boron compounds, and new therapeutic strategies that further improve the efficacy and safety of BNCT.

Our goal is not only to generate new scientific knowledge, but also to answer fundamental clinical questions: Why does BNCT work? Why does it fail in some cases? How can it be made safer and more effective for a broader range of patients? By addressing these questions, we aim to translate our discoveries into clinical practice.

For us, basic research is more than laboratory science—it is the blueprint for tomorrow's cancer treatments. It lies at the heart of the mission of the Fujita Health University BNCT Research Center, providing the scientific foundation for innovations that will ultimately benefit patients worldwide.
